Title: Enantiomer-specific isotope analysis (ESIA) of D-and L-alanine: nitrogen isotopic hetero-and homogeneity by microbial process and chemical process. Abstract Nitrogen isotopic hetero-and homogeneity of D-α-alanine and L-α-alanine were investigated in microbial processes in the domain Bacteria and in chemical processes in symmetric organic synthesis. D-alanine is an enantiomer that is physiologically essential for microbial growth and metabolic maintenance. The nitrogen isotopic difference Δ 15 N D-L 20 (defined as δ 15 N D-Ala − δ 15 N L-Ala) in peptidoglycan amino acids in bacteria such as the representative gram-positive phyla Firmicutes and Actinobacteria (Enterococcus faecalis, Staphylococcus aureus, Staphylococcus staphylolyticus, Lactobacillus acidophilus, Bacillus subtilis, Micrococcus luteus, and Streptomyces sp.) tended to be 15 N-depleted in D-alanine (Δ 15 N D-L < −2.0‰). These results suggest that the composition of isotopically heterogeneous 25 components in these bacteria is primarily controlled by enzymatic pathways prior to formation of the bacterial cell wall. In contrast, the Δ 15 N D-L of racemic alanine in the chemical pathway during the nucleophilic substitution reaction (SN1 type) between 2-bromopropionic acid and ammonia identified fully homogeneous components for each enantiomer. The novel enantiomer-specific isotopic analysis (ESIA) method is useful in 30 determining the origins of chirality in biogenic and abiogenic processes and is applicable to enantiomer studies.

Joint interpretation of enantiomer and stable isotope fractionation for chiral pesticides degradation.

Chiral pesticides are important contaminants affecting the health and functioning of aquatic systems. The combination of stable isotope and enantiomer analysis techniques has been recently proposed to better characterize the fate of these contaminants in natural and engineered settings. We introduce a modeling approach with the aim of unifying and integrating the interpretation of isotopic and ...

Abiogenic and biogenic chiral amino acids for further enantiomer-specific isotope analysis (ESIA)

Introduction: Since the time of Pasteur, the development of biomolecular chirality has remained one of the most important problems with regard to our knowledge concerning chemical evolution. The homochiral amino acids and sugars are essential to the formation, structure, and function of biopolymers and are a defining molecular trait of life on the Earth [1] (cf. asymmetric photochemical process...

Ab initio study of nitrogen and position-specific oxygen kinetic isotope effects in the NO + O3 reaction.

Ab initio calculations have been carried out to investigate nitrogen (k15/k14) and position-specific oxygen (k17/k16O & k18/k16) kinetic isotope effects (KIEs) for the reaction between NO and O3 using CCSD(T)/6-31G(d) and CCSD(T)/6-311G(d) derived frequencies in the complete Bigeleisen equations. Isotopic enrichment factors are calculated to be -6.7‰, -1.3‰, -44.7‰, -14.1‰, and -0.3‰ at 298 K f...

Insights on the marine microbial nitrogen cycle from isotopic approaches to nitrification

The microbial nitrogen (N) cycle involves a variety of redox processes that control the availability and speciation of N in the environment and that are involved with the production of nitrous oxide (N(2)O), a climatically important greenhouse gas. Isotopic measurements of ammonium (NH(+) (4)), nitrite (NO(-) (2)), nitrate (NO(-) (3)), and N(2)O can now be used to track the cycling of these com...
